چکیده
A clickable, affinity-based probe (AfBP), which was modified from staurosporine (a natural product kinase inhibitor), has been synthesized and used in situ for activity-based proteome profiling of potential cellular targets of staurosporine in HepG2 cancer cells.

A tuned affinity-based staurosporine probe for in situ profiling of protein kinases.
A clickable and cell-permeable affinity-based probe (AfBP) was designed from staurosporine, by incorporating an electrophilic chloroacetamide warhead to facilitate in situ proteome labeling of a range of potential kinase targets.
متن کاملTarget profiling of zerumbone using a novel cell-permeable clickable probe and quantitative chemical proteomics.
Zerumbone is a phytochemical with diverse biological activities ranging from anti-inflammatory to anti-cancer properties; however, to date the cellular targets of this important compound have remained elusive. Here we report the global protein target spectrum of zerumbone in living cancer cells using competitive activity-based protein profiling of a novel cell-permeable clickable probe, combine...
متن کاملMultiplex Imaging and Cellular Target Identification of Kinase Inhibitors via an Affinity-Based Proteome Profiling Approach
MLN8237 is a highly potent and presumably selective inhibitor of Aurora kinase A (AKA) and has shown promising antitumor activities. Like other kinase inhibitors which target the ATP-binding site of kinases, MLN8237 might be expected to have potential cellular off-targets. Herein, we report the first photoaffinity-based, small molecule AKA probe capable of both live-cell imaging of AKA activiti...
متن کاملIn cell scalaradial interactome profiling using a bio-orthogonal clickable probe.
A bio-orthogonal click-chemistry procedure was developed to allow the in cell interactome profiling of scalaradial, an anti-inflammatory marine natural product. The results were validated through the application of the classical in vitro chemical proteomics and several bio-physical methods; peroxiredoxins, 14-3-3 isoforms and proteasomes were recognized as main scalaradial targets.
متن کاملMining the thiol proteome for sulfenic acid modifications reveals new targets for oxidation in cells.
Oxidation of cysteine to sulfenic acid has emerged as a biologically relevant post-translational modification with particular importance in redox-mediated signal transduction; however, the identity of modified proteins remains largely unknown. We recently reported DAz-1, a cell-permeable chemical probe capable of detecting sulfenic acid modified proteins directly in living cells. Here we descri...
